Abstract:
This article explores the role of mitochondrial functional homeostasis and immunogenic cell death in the occurrence and development of liver injury, as well as the hepatoprotective effects of traditional Chinese medicine (TCM) targeting the mitochondrial homeostasis-immunogenic cell death network. Factors such as oxidative stress, energy metabolism disorders, mitochondrial membrane potential disturbances, and mitochondrial DNA damage can lead to an imbalance in mitochondrial functional homeostasis, which in turn mediates inflammatory injury of hepatocytes through various signaling pathways, ultimately resulting in immunogenic cell death. This indicates that mitochondrial functional homeostasis plays an important role in cellular immunogenic cell death. Natural medicines can exert hepatoprotective effects by targeting mitochondrial membrane potential, inhibiting mitochondrial respiratory chain complexes, interfering with mitochondrial metabolism, regulating mitochondrial reactive oxygen species (ROS) levels, and influencing mitophagy processes. Therefore, studying the relationship among mitochondrial functional homeostasis, immunogenic cell death, and liver injury, as well as the interventional effects of natural medicines, is of great value for gaining a deeper understanding of novel pathogenic mechanisms of liver injury and for developing new therapeutic strategies.

摘要:本文探讨线粒体功能稳态、免疫原性细胞死亡在肝损伤发生发展中的作用及中医药靶向干预线粒体稳态-免疫原性细胞死亡网络的肝保护效应。氧化应激、能量代谢障碍、线粒体膜电位紊乱、线粒体DNA损伤等因素会引起线粒体功能稳态失衡,进而通过多种信号通路介导肝细胞炎性损伤,最终引起肝细胞免疫原性死亡发生,说明线粒体功能稳态在细胞免疫原性死亡中发挥着重要作用。天然药物可通过靶向线粒体膜电位、抑制线粒体呼吸链复合物、干扰线粒体代谢、调节线粒体活性氧(ROS)水平以及影响线粒体自噬过程等途径发挥肝保护作用。因此,研究线粒体功能稳态、免疫原性细胞死亡与肝损伤的关系以及天然药物的干预作用,对于深入了解肝损伤的新发病机制、研发新型治疗策略具有重要价值。
